Frankly, it's difficult to understand what you are really looking for.
The short answer would be: "Yes, it is possible.", but how to achieve it is whole different ball-game. Realistic? Cartoonish?
If you want to hand-paint everything I'd recommend SP, if you want to use bakes such as AO or Curvature maps, you can mix procedural noises with colored versions of your bakes like explained in this tutorial:

You can also use photos, process them in SD, and use them as stencils in SP.

The plugin path seems to be wrong as the Substance plugin is now shipping as a module (since 2013.5 if I am not mistaken...).
Did you upgrade from 2012 to 2015? Looks like a preference issue to me, try to unload and reload the plugin manually in Plugin Manager.

You're missing the bitmap baking stage: Select your Substance Node and under "Automatic Baking" check "Automatic Connections" et re-create your shader network.

Well, I guess you have to map and adapt your outputs depending on your needs. Roughness and Metallic output usage have been introduced in SD to match the Physically Based Rendering pipeline, which is commonly used in the Games industry now.
But it doesn't mean that you have to stick with this workflow.

I haven't used Vray for a while, but as far as I remember all shaders (including the default VrayMtl) supports this kind of material:

Are you planning to animate those textures at render time?

Only the "Help" menu does not work on Maya 2014 (unless you have a compiled version of PyQt for Maya 2014). All other scripts are working fine.
I am still working on an update version for 2014+, there is a bit of work though as I need to switch every module to PySide. I cannot give any ETA for this, sorry.

If you simply need bitmaps you can use the "Export images to disk" function.

Can you try to execute this in a Python shell within Script Editor? :

PS: Select the substance node first before executing the script.

As far as I remember, this was a known issue in 3.x, fixed since ver.4.
you should upgrade to the latest version of Unity and see if it happens again.
