I want to export to other software to support the (RGBA)Channel..But i found the spexp file too hardly understand ....

Export presets are not configuration files that are meant to be human readable, they are binary files in our own format. You will have to use Substance Painter to edit them properly.
Don't forget your log file. It can be exported from the Help menu of the software.

Would there, actually, be a reader to the export presets?

I would like to access the information from the output maps and input maps on the configuration tab of the Export Textures tool. If there is no export preset reader, how to access those through scripting?

It's not possible at the moment, the API doesn't provide function to query the export configuration in such details.
Don't forget your log file. It can be exported from the Help menu of the software.
